that mice deficient in calbindin-D 28K exhibit cerebellar ataxia (Airaksinen et al., 1997a) and impaired spatial Hongo, Bunkyo-ku Tokyo 113 learning (Molinari et al., 1996). However, the properties of high-affinity Ca 2ϩ buffering in vivo and its specific Japan † Department of Pharmacology and Physiology consequences for neuronal Ca 2ϩ signaling have not been elucidated. We have therefore now investigated Ca 2ϩ buffering in Purkinje cells with the use of a low-affinity (‫01ف‬ ␮M) Ca 2ϩ indicator dye (BTC) and a high-affinity (19 nM) caged Ca 2ϩ compound (DMNPE-4). Neither of these Japan compounds interfered to a substantial extent with the intrinsic Ca 2ϩ-buffering properties of these cells. We detected the existence of a large concentration of a high-Summary affinity, cooperative, and mobile Ca 2ϩ buffer, the properties of which resemble those of calbindin-D 28K. Endogenous high-affinity Ca2+ buffering and its roles were investigated in mouse cerebellar Purkinje cells with the use of a low-affinity Ca2+ indicator and a high-affinity caged Ca2+ compound. Increases in the cytosolic Ca2+ concentration ([Ca2+]i) were markedly facilitated during repetitive depolarization, resulting in the generation of steep micromolar Ca2+ gradients along dendrites. Spontaneous ‘mini’ release occurs at all synapses, but its nature remains enigmatic. We found that > 95% of spontaneous release in murine cortical neurons was induced by Ca21-binding to synaptotagmin-1 (Syt1), the Ca21 sensor for fast synchronous neurotransmitter release. Thus, spontaneous and evoked release used the same Ca21-dependent release mechanism. As a consequence, Syt1 mutations that a...
